RESUMO

Oligosaccharides are low molecular weight carbohydrates synthesised by living organisms, related with immune defence, viral replication, cell growth, cellcell adhesion and energy storage. Furthermore, they are considered functional foods as a result of their prebiotics properties, which stimulate the intestinal micro flora improving the organic physiology. In this work, the production of oligosaccharides by Zymomonas mobilis strain ZAG-12 was studied. Fermentations in sucrose medium containing yeast extract and mineral salts were developed based on previous experiments using factorial method for optimisation. Analyses of the produced oligosaccharides by High Performance Liquid Chromatography (HPLC) were done. Additional experiments in flasks at 35 and 40oC sucrose 250 g/L, at 100 rpm of agitation were developed. The pH, biomass, levan and oligosaccharides produced and sucrose consumed were analysed. Finally, a kinetic study in a bioreactor Bioflow III (New Brunswick), using medium with sucrose 150 g/L, at 30 oC, 100 rpm, in batch fermentation process, during 72 h was carry out. The levan production was considered low at 35 and 40 oC, possibly due to by-products formation including oligosaccharides. Oligosaccharides with polymerisation degrees (DP) between 3 and 15 at 35 and 40 oC was detected. The predominating DP were 3,5,6,12,13 and 15 at these temperatures. In the production at 30oC it was observed oligosaccharides with DP between 4-15, with higher production of 4, 5, 6, 7, 8 e 15 degrees. The results show that Zymomonas mobilis strain ZAG-12 is a potential oligosaccharide producer and that 30, 35 e 40oC are better temperatures for oligosaccharide production. Oligosaccharides with low degree of polymerisation are formed mainly at 40oC, but the best yield was observed at 30 oC
