Efeito da lectina de Canavalia brasiliensis sobre a expressÃo e atividade de metaloproteinases extraÃdas de lesÃes cutÃneas de camudongos

RESUMO

This study presents results from the Canavalia brasilie lectin use of nsis seeds, on the expression and metalloproteinases activity extracted from skin experimental lesions. The lectin effect was tested in four therapeutic schemes (2nd, 7th and 12th days after surgery), according to the following treatment: (10μg) ConBr, 150mM NaCl, (10μg) ConBr/mannose prepared in physiologic solution with 0.1M mannose and 0.1M mannose. The study proceeded with the collection of surgical lesions for the injury area analyses, histopathology, colagenolytic activity using azocoll and real-time PCR with specific primers for metalloproteinase 2 (MMP-2) and metalloproteinase 9 (MMP-9). Treated ConBr group showed inflammatory signs less intense and greater wound closure when compared to the other groups. Colagenolytic activity was present in all groups studied, but the ConBr group had the lowest activity on the 2nd day after surgery, however the ConBr/mannose group obtained the highest activity at the mentioned time above, this profile has remained for the ConBr/mannose group in the 12th day after surgery. This activity profile may be related to the histopathology findings, which showed a similar pattern in the organization tissue between ConBr and ConBr/mannose groups, but the latter showed a better organization of the collagen fibers. It was observed that injuries area presented a statistically significant difference between the ConBr group and the control group (NaCl) on the 7th and 12th day after surgery. The MMP-2 and MMP-9 gene expression was observed at all times of study for ConBr, ConBr/mannose and mannose groups, with different quantities, but the NaCl group did not expressed MMP-2 and MMP-9 gene in 2nd day after surgery and the MMP-9 gene in 12th day. Despite the ConBr/mannose group had presented a better organization of collagen fibers, the ConBr group showed positive for the wound closure in comparation to other groups
