Diterpenos de Myrocarpus frondosus Allemão : isolamento, determinação estrutural e atividade biologica

RESUMO

The phytochemical study of hexane and dichloromethane extracts trom the roots ot Myrocarpus frondosus resulted in the isolation of seven diterpenes (18, 19, 20, 21, 22, 23 and 24) whose strutuctures were determined by the analysis of mass spectra (Iow and high resolution and EM/EM experiments), 1D-NMR (H, C, DEPT e NOESY 1 D) and 2D-NMR (gCOSY, HSQC/HETCOR e HMBC/COLOC). A bibliografic research on Chemical Abstract (1907-2004) showed that 19, 20, 21 and 22 are new compounds. The hexane, dichlorometane and methanol extracts from the roots were active when submitted to bioassay Artemia salina lethality. The extracts and compounds 18, 19 and 21 were submitted to bioautography assay, when the hexane extract showed antimicrobial activity against Rhizopus oryzae (CCT 4964) and Alternaria alternata (CCT 1250), while 19 it was active against Bacillus subtilis (CCT 0089) and Staphylococcus aureus (CCT 4295), both giving MIC = 1000 mg mL. The hexane and dichloromethane extracts were submitted to the test of antiproriferative activity against lineages of tumoral cells from humans and showed citostatic and citocidi effect, while the methanol extract showed only citostatic effect. The compound 21 showed activity against OVCAR (ovarian) and PCO.3 (prostate) lineages, being about 1000 and 60 times more active than doxorubicin (positive control) respectively, while the activity against NCI.460 (lung) lineage was equivalent to that of the positive control (doxorubicin).
