Estudo fitoquimico de Poecilanthe parviflora Benth. e Lonchocarpus atropurpureus Benth. (Leguminosae) isolamento, determinação estrutural e atividade biologica

RESUMO

Dichloromethanic extract from Poecilanthe parviflora roots was submitted to chromatographic analysis and furnished a mixture of steroids ( campesterol 1, b-sitosterol 2 and stigmasterol 3) and an unsaturated fatty acid methyl ester(C-22, three unsaturations) 6. Acetylation of methanolic extract followed by chromatographic analysis furnished peracetylated sucrose 4 and quebrachitol 5. Light petroleum extract from Lonchocarpus atropurpureus roots was submitted to chromatographic analysis allowing the isolation of five flavonoids (7, 8, 9, 10 and 11) and a mixture consisted by two pentaciclic tritepenes (12 and 13). Minimiflorin 7, mundulinol 10 and mundulin 11 have been isolated before from other sources while 5,2 -dihydroxy-3-methoxy-6,7-(2",2"-dimethylcromene)-8-(3" ,3" -dimethylallyl)-flavanone 8 and 3, 5-dihydroxy-6,7-(2",2"-dimethylcromene)-8- (3" ,3" -dimethylallyl)-2 -hydroxy-flavanone 9 are new compounds. The two pentaciclic triterpenes were determinate to be b-amirin 12 and lupeol 13.The molecular structures were elucidated by means of their respective spectral data (UV, IR, MS, H NMR, C NMR) and bidimensional spectra (HETCOR, HSQC, HMBC). Flavonoids 7, 8 and 9 present an OH on C-2 which is not a very frequent substitution position. Quantitative analysis of light petroleum extract of L.atropurpureus roots by using HPLC reversed phase showed that 8 is the most abundant flavonoid. The roots s extracts of P.parviflora were tested against some fungi and bacteria and the roots s extracts of L.atropupureus were tested against some fungi through the bioautigraphy method and all were inactive. The extracts of L.atropurpureus roots and the new flavonoids 8 and 9 showed activity in the Brine Shrimp bioassay. Flavonoid 8 showed no activity when it was tested against three cancer lineage (citotoxicity in vitro) but showed a light increase in the insulinic activity when it was tested in a model in vivo.
