RESUMO

Failures, faced in schitossomiasis treatment with prazinquantel, are signing to the necessity to search new drugs to the treatment of this disease as well as monitoring its activity against strains of Schistosoma. The objective of the present work is to evaluate the schistossomicide activity in vivo of the Prazinquantel (PZQ) and the imidazolidine derived 3-(4-chlorobenzyl)-5-(4-nitrobenzylidene)-imidazolidin-2,4-dione (FZ-4). It was used nine groups of Swiss lab rats with six animals each group, infected experimentally with 80 S. mansoni parasites (BH strain), and treated orally during five days with 25, 50 e 100 mg/Kg, 50 days after the infection. A control group was used to the PZQ and another to the FZ-4. The prazinquantel showed to be efficient in reducing the parasite load in infected animals, reducing in 100% on doses of 100 and 200mg/Kg. The calculated ED50 was 14,75mg/Kg, demonstrating that the BH strain of S. mansoni was sensible to Praziquantel. The FZ-4, in doses used in the present study, did not show capable of reducing the number of verms nor either alter the oogram in infected animals, however, with the 25mg/Kg dose showed a significant raise in the number of eggs per fezes grams, when analyzed in the sixth and fourteenth day after the end of the treatment. The histopatologic analysis indicated that the FZ-4 reduced the areas of fibrosis and protection of the intestinal mucosa in the granulomatosic reaction, activating local linfatic nodules with the 100mg/Kg dosis. Concluding the present study, the activity against S. mansoni was observed, but no activity was observed when FZ-4 was used, however parasite eggs reduction in fezes was observed
