RESUMO

Biomphalaria glabrata is the most susceptible intermediary host to Schistosoma mansoni in Brazil. The use of plants with molluscicidal properties could be an approach to local control of a vector. As alternative test to show chemic and natural products toxicity has been used Artemia salina, an invertebrate with industry and aquiculture importance. Cratylia mollis is a native plant from Semiarid Region of the Northeast of Pernambuco. The presence of lectins in Cratylia mollis seeds was available .Lectins are proteins or glycoprotein with the capacity of binding specifically and reversibly to carbohydrates. Three isolectins were obtained from different fractions (F) from saline precipitated (F0-40, F40- 60, F60-80). Cramoll 1,4 was obtained from F40-60, purified by affinity chromatograph. Cramoll 1,4 and F60-80 promoted death of all analyzed Biomphalaria glabrata specimen death with 80 and 100 ppm. Cramoll 1,4 (LC90 = 23.581) showed more toxicity against A. salina than F60-80 (LC90 = 62.988), but neither of them had activity with its egg mass and cercaria. The toxicity to A. salina is an indicative of potential use of Cramoll 1,4 and F60-80 as inhibitors of tumor cell growth. Therefore, from these results the effect of Cramoll 1,4 on Walker AR (rat sarcoma) and LNCaP tumor cells line, originally obtained from metastatic human prostate carcinoma, was investigated. Cramoll 1,4 in low concentration (4 μg/ml) induced apoptosis on both tumor cell line, Walker AR tumor and LNCaP with 4 hours of treatment. Analyses realized in 2.5 h of treatment promoted increase in cytosolic Ca2+ but this effect was not sawn in mitochondria. Cramoll 1,4 also increased reactive oxygen species (ROS) production, which was reduced in presence of DPI suggesting that ROS production occur on cytosol. Additional tests realized with walker tumor cells showed that Cramoll 1,4 antitumoral activity was totally inhibited by antioxidant desferal (DFO) and partially inhibited by inhibitor of Caspase 8. This present works results demonstrated that Cramoll 1,4 has potential to be used: 1) on B. glabrata control; 2) As promising compound on alternative anticancer therapy, since induce tumor cells death through apoptosis resulting in a structural and vital functional irreversible lost of these cells; 3) Laboratorial diagnosing of abnormal glycoprotein from dengue patients serum.
