Perfil FarmacolÃgico e ToxicolÃgico de Produtos Bioativos Obtidos de Streptomyces spp Isolados do Leito de Rios do Estado da ParaÃba

RESUMO

Aimed to search for species of Streptomyces isolated from mesoregions of Paraiba producers of bioactive metabolites with pharmacological activity, assessing the levels of toxicity of extracts and their chemical composition. 49 species of the genus Streptomyces were isolated from 68 samples of soil bed in the main rivers in different mesoregiÃes the state of Paraiba-Brazil. The Streptomyces were obtained through the suspension of soil in saline (0.89%) and striated in the Petri dish. The identification was done through analysis macromorfologic, micromorphologic and physiological tests. The antimicrobial screening was determined by the technique in blocks of agar, and the dried ethanolic extracts prepared from the species that showed the best results, respectively strains SP1 and SP3. The evaluation of the antagonism was performed through the techniques of disc diffusion in solid medium, microdilution and determination of the curve of microbial death. Tests of anti-inflammatory activity, acute toxicity and antitumour were conducted under specific literature, using animal models previously approved in Committee. The chemical characterization was done by chromatographic and spectroscopic tests. The results characterize a potential antimicrobial extracts of sp-1 and sp-3 in front of the yeast species of the genus Candida spp., genus Aspergillus sp., gender and Trichophyton sp samples of Staphylococcus spp in clinical and home collection. The tumor inhibition was observed in both extracts against the tumor Sarcoma 180 with up to 73% reduction in the weight of the tumor mass. There was no apparent anti-inflammatory action of extracts sp-1 and sp-3. The extracts showed effects of SNC stimulants and depressants in the first hours of the continuing acute toxicity test, showing no deaths, so it is not possible to characterize the lethal dose for each statement. The chemical characterization showed the extract from the sp-1, the meeting of 13 fractions, which was evidenced after recrystallization the compound sp-1/09. Besides the other fractions are at the stage of purification for elucidation. Become timely and rewarding the performance of new samples of soil from various regions paraibanas revealing the study of Streptomyces spp. biotechnology as an important source to obtain new bioactive compounds
