Padronização e processo de invasão/inibição de esporozoitos de Eimeria acervulina e E. tenella em celulas primarias de galinha, na presença de lectinas de plantas

RESUMO

The effect of six plant lectins in the invasion of Eimeria acervulina and E. tenella sporozoites using primary chick kidney and embryo cells cultivated in vitro, was examined. Pretreatment of sporozoites with l00 ?g/ml of DViol, DVir, DGr, ConBr, CF, PP and 50 ?g/ml of DViol, DVir, ConBr and PP inhibited significantly the invasion of E. acervulina and E. tenella sporozoites, respectively. Pretreatment of cell cultures with l00 ?g/ml l of lectins had no significant effect on E. acervulina sporozoites invasion. There was no correlation between sugar affinity of lectins for D-manose and the inhibitory effect of the parasites demonstrated by treatment of E. acervulina and E. tenella sporozoites with DViol and incubated with or without D-manose which showed similar inhibitory effects. Flow citometric analysis of E. acervu/ina sporozoites treated with l00 ?g/ml of six lectins showed no membrane disruption and loss of viability. The data observed in the present study suggest that specific interactions between Diocleinae lectins and E. acervu/ina and E. tenella sporozoites can change the process of invasion in cells cultivated in vitro without showing parasiticide activity. The experiments carried out showed higher effect of lectins on E. acervulina sporozoites than for E. tenella sporozoites
