Isolamento e seleção de microrganismos brasileiros para reações de biocatalise e produção de metabolitos

RESUMO

This thesis is a pioneer work at the Chemistry Institute/UNICAMP, consisting of a preliminary investigation of a Brazilian Culture Collection biocatalitic potential (Coleção de Culturas Tropical ,CCT, Fundação Tropical de Pesquisas "André Tosello") 13 different microorganisms from the above mentioned collection were evaluated: fungi (Aspergillus niger CCT 2760, Aspergillus niger CCT 3086, Aspergillus niger CCT 4846, Aspergillus oryzae CCT 0975, Aspergillus terreus CCT 3320, Aspergillus terreus CCT 4083, Curvularia eragrostidis CCT 5634, Emericella nidulans CCT 3119, Geotrichum candidum CCT 1205, Phanerochaete chrysosporium CCT 1999 and Rhizopus oryzae CCT 4964), bacteria (Corynebacterium aquaticum CCT 3023) and yeast (Rhodotorurla glutinis CCT 2182). The detected enzymatic activities separated by reactions were sulfide oxidations, Baeyer-ViIIiger, hydroxylation, ketone reductions, ester hydrolyses (and hydrolyses epoxide. Taking into account the enantioselectivity of the reaction the best results were the production of (-)-(S)-phenyl-ethyl-sulfoxide (ee>99%), phenyl-methyl-sultoxide (ee>99%), p-methyl-phenyl-ethyl-sulfoxide (ee>95%), (-)-(S)-p-methyl-phenyl-ethyl-sulfoxide (ee>99%), m-chloro-phenyl-ethyl-sulfoxide (ee>99%), (+)-(R)-5-methyl-heptalactone (ee 96%), cis-(+)-(1R,2S)-2-methyl-cyclehexan-1-ol (ee >95%), cis-(+)-(2S,3S)-2-methyl-2-allyl-3 hydroxy-cyclehexan-1-one (ee>98%), (-)-(S)-2-acetoxy-methylbenzyl (ee 96%), (+)-(R)-2-hydroxyl-mehylbenzyl (ee 83%), 4-acetoxy-3-methyl-cyclehex-2-en-1-one (ee >98%), (+)-(R)-1.2-epoxy-octane (ee 69%) and (-)-(S)-1,2-octanediol (ee 80%). In addition two novel secondary metabolites 3,6-dihydroxy-5-methyl-1-oxospyro[benzopiran-10-tetrahydrofurane] and the 5,5,12-trimethyl-3,7-isopropoxy-pentane were isolated from Aspergillus terreus CCT 3320 and Aspergillus niger CCT 1435, respectively. I should be mentioned that as a training program, 31 bacteria from Trigona bees were isolated and deposited at the CCT for future research.
