Efeitos genotóxicos e mutagênicos da fração solúvel do óleo diesel em uma espécie de peixe Neotropical

RESUMO

ABSTRACT The present work shows results of the mutagenicity and genotoxicity evaluation of the comercial diesel oil fraction on freshwater fishes Prochilodus lineatus (Characiformes: Prochilodontidae), ordinarily known as curimba. The water containing the diesel oil soluble fraction was obtained through simulation of the spill in tropical environment. In the spill, the oil has been mixed with water in glasses aquariums (50L), exposed to intense sun light followed by collecting the diesel water soluble fraction on opaque gallons and kept until the moment of the experiences. The animals were exposed to the diesel water soluble fraction (DWSF) diluted on 50% with artesian well water, in acute way (6, 24 and 96 hours) and sub-chronic way (15 days) on 100L aquarium containing eight fishes each. Artesian well water has been employed as negative ? control. The positive ? control was performed with animals wich received ciclophosphamide injection. After exposurings, the blood from the caudal vein of the animals were collected to achieve the Comet and Micronuclei assays. In the statistics analyses Mann-Whitney and Kruskal-Wallis non-parametrics tests have been used. By the Comet assay all animals exposed to the diesel water soluble fraction presented an increase on damaged cells frequency in comparison to theirs respective negative ? controls. As well as this the score of cell damage was statistical differently from those observed on the animals exposed only in water from artesian well. The lesions caused in the genetic material of fishes exposed to DWSF might be resulted from oxidative damages by eletrophilics compounds originated on metabolized diesel oil hydrocarbons. In every exposures to DWSF a crease on the frequency of micronucleus on erythrocytes of exposed fishes was in op tested. Therefore, the combination of both methodologies at the present work has shown suitable and profitable in evaluating genotoxicity of the diesel oil water soluble fraction due to its complementarities. Consequently, the present work that the diesel oil spill on tropical environment may carry, in short or medium term, damages on the genetic material of this Brazilian neotropical fish specie.
