Efeitos do extrato liofilizado da Euphorbia tirucalli sobre a resposta hematopoetica em camundongos portadores do tumor ascistico de Ehrlich

RESUMO

In this work, we have investigated the effects of the liofilizated extract of the Euphorbia tiruca1li(TEE), onthe growthand differentiation of hematopoietic progenitors for granuiocyte/macrophage (CFU-GM) in the bone marrOWanelspieen of Ehrlich ascites tumor-bearing mice (EAT). !vWeBaib/c mice were inoculated intraperitoneaily (i.p.) with 6xI06 viabie tumorcells/mouse and 125,250 or 500mg/Kg ofTEE were given orai1yfor 5 consecutive days. Thebone marrow and spieen progenitorceUs were coUected 24 hours afier the Iast dose of TEE. Afier that we studied the growth anel differeniiationofcolony forming unit (CFIJ-G!vI),through the technique of cionai culture of hematopoietic ceUs in semi-solidmedium. Intumor bearing-mice, the total number of CFtT-G!vlper femur was significantiy redu~ed whereas in the spieen the number of CFU-G!vl was dramaticaily increased. Tne treatmentof these animais withthree doses of TEE reverted the meduUar supression anel reduced the increased extrameduUar hematopoiese, which was induced bythe tumor. Tne vaiues found for the animalsthat were treat~ with threediffernts doses of TEE orny, stayed near the rates of normalanimals. The survival rate ofthe aniIIijl!swas evaluat~ for 29daysafier inocuiation ofEAT. We verified that treatigíhese animaIs with three ,dosesof TEE, e~nded significantly their survival rate ftom 21 to 27.and29 days, depending thedoseofiúthat wasuseçl. The resulis demonstrated thattreatuleni with TEE produced proteçtive effects on the myelopoietic response of tumor~bearing mice.
