RESUMO

Chagas disease continues to be a significant public health problem. The only available treatment, benznidazole, causes serious side effects and have therapeutic limitations. Aiming to search alternative treatment to this disease, in the present study was analyzed the in vitro effects of acetone extract and their isolated components fumarprotocetrÃric (FUM) and protocetraric (PRO) acids from Cladonia verticillaris (Raddi) Fr. against Trypanosoma cruzi, in vitro. For obtetion of acetonic extract were submitted 100g C. verticillaris an eluotropic series of organic solvents. After obtention of this extract and their isolated components the (FUM) and (PRO). The bioassays were carried out with epimastigotes and bloodstream tripomastigotes and this forms were incubated during 72 and 24 hours respectively in the absence or presence of 75 to 300ig.mL-1of test compounds. The IC50 and IL50 was estimated directly by cell counting. The assays were carried out in triplicates, in three independent experiments. The treatment of epimastigotes forms resulted in cells growth inhibition, in a dose dependent way. The inhibition degree was different between the compounds. The trypomastigotes forms exhibited higher susceptibility to C. verticillaris secondary metabolites, with 100% of parasites lysis at 300 igÂmL-1 of PRO. Ultrastructural analysis trypomastigote forms treated with PRO showed organelle disorganization, nuclear retraction and kinetoplast and mitochondrial demage. Take together, the obtained results stimulate further studies in order to elucidate the mechanism of FUM and PRO action, as well as to evaluated their effects on intracellular amastigotes forms. The results also point towards the PRO as potential agent against Trypanosoma cruzi.
