RESUMO
A total of 803 yeast isolates were obtained from quarterly collected samples from fermented must from four different fuel-ethanol distilleries located in Northern Brasil: 110 isolates from Miriri distillery, 90 isolates from Giasa distillery and 443 isolates from Japungu distillery, all from State of ParaÃba, and 160 isolates from JB distillery located at the State of Pernambuco. All yeast isolates were subjected to molecular fingerprinting analysis by using the PCR-based marker (GTG)5 primer, which allow the amplification of inter simple sequence repeats between two microsatellites in the yeast DNA. In parallel, DNA samples from S. cerevisae type strains, different species of the Saccharomyces sensu stricto and non-Saccharomyces species were analysed. The results showed that all S. cerevisiae type and industrial strains produced amplification patterns that shared common bands not present in other Saccharomyces species, neither in the non-Saccharomyce species tested. It was demonstrated that (GTG)5 can be used to discriminate S. cerevisiae from any other yeast species, which suggest it use for further molecular taxonomy. Moreover, polimorphic amplified bands were identified among many S. cerevisiae strains, both type and industrial isolates, which allowed the discrimination of 17 yeast strains. Taken together, the results obtained in this work demonstrated that (GTG)5 primer can be promptly used for monitoring populations in the fermentation process. The isolates belonging to P1 and P18 (GTG)5 amplification patterns showed the best fermentative capcity among all isolates tested, as well as high persistence in the fermentative process. Therefore, they are candidates for further genetic manipulation of industrial interest
