Avaliação de metodos bioquimicos e sorologicos na caracterização de especies de Candida isoladas da cavidade bucal de humanos

RESUMO

This work had the purpose to evaluate biochemistry and serological methods to characterize Candida species from human oral cavity, emphasizing taxonomy, systematic and epidemiological studies. The strains used represent the five Candida species: 97-a, F-72, E-37, 17-b and CBS-562T (C. albicans), FCF-405 and FCF-152 and CBS-566T (C. guilliermondii), 21-c, 7-a and CBS604T (C. parapsilosis), IM-90, 4-c and CBS-573T (C. krusei), l-b, FCF-430, Ct-4 and CBS-94T (C. tropicalis). As negative control the strain IZ-1339 (Kluyveromyces marxianus) were used. The biochemical and serological methods used were based upon the growth in cromogenic medium CHROMagar Candidaâ, ELISA (enzyme-linked immunosorbent assay) and protein electrophoresis (SDS-PAGE). The method CHROMagar Candidaâ showed specificity for the C. albicans, C. tropicalis and C. krusei species but it was not able to identify the C. guilliermondii e C. parapsilosis species, since it showed rose and vanilla color, respectively, which is not specific for these two species. The SDS-PAGE showed significative differences in the electrophoretic pattems from the proteic cellular extracts, showing a decrease of the number of bands compared to the increase of the centrifugation time. The ELISA could not present differences among the cell extracts, expressed in O.D., when compared. In an increasing order of specificity, the SDS-PAGE seemes to be the more efficient for the strains characterization being able to group the species, followed by the CHROMagar Candidaâ which showed specificity for the C. albicans, C. tropicalis and C. krusei, but not for the remaining species studied. The ELISA using polyclonal antibodies was ineflective to identify species due to the high level of cross-reaction
