Análise proteômica comparativa entre neutrófilos não ativados e ativados com PMA, um análogo do diacilglicerol

RESUMO

Neutrophils are the major type of leukocytes in peripheral blood and protect against fungal and bacterial infections. Directed migration in a gradient of chemotactic stimuli enables these cells to rapidly find the site of infection and destroy the invading pathogens. PMNs can exist in three different activation states, namely, resting, primed, and activated states. Activated neutrophils produce reactive oxygen species (ROS), in what is known as respiratory burst, increase phagocytosis, delay apoptosis, among other functions. These cells can also be activated in vitro by particles and substances such as phorbol-myrystate-acetate (PMA). The PMA is a diacylglicerol (DAG) analog that bypasses membrane receptors and directly activates intracellular protein kinase C isoforms (PKC). In this study, human neutrophils were prepared from peripheral venous blood of healthy volunteers (greater than 96% purity, greater than 97% viability) and were obtained by density centrifugation on percoll gradient. 107 cells were activated with 100ng PMA for 30 minutes in 1 mL of PBS. Cytometric analysis was executed for measurement of respiratory burst, viability and purity from neutrophils. A comparative analysis of 2D-eletrophoresis gels, of normal and PMA activated neutrophils was executed, and 83 spots revealed differential expression, from which 13 spots were up regulated after PMA activation and 70 were down regulated in this condition (p≤0,05). Moreover, 141 spots were regarded as belonging only to normal gel, and 109 spots were exclusive of the PMA activated neutrophil map Thirty five conserved spots were subjected to peptide mass fingerprinting, yielding six identifications. Among the identified proteins, four are related to the inflammatory process and two are classified as structural. RBCK1 and CRAa, were found for first time in neutrophils. Our hypothesis is that MEK1 and MEK kinase 1 (MEKK1) represses the RBCK1 activity, and PKA enhances its activity. Lysozyme, a microbicidal protein of specific granules, and β-actin, the main isoform of neutrophil cytoskeleton were identified too. MRP-14, present in the neutrophil cytoplasm, provides bacteriostatic and cytokine-like effects.
