1. Presence of ascites in bullfrog breeders reared and kept on a frog farm

Chapters one and two deal with the isolation and identification of bioactive peptides from the skin secretion of Phyllomedusa hypochondrialis. The first chapter demonstrates the use of protemic techniques, mainly mass spectrometry, for the study of bradykinin related peptides (BRPs) present in the amphibians secretion and freshly dissected skin fragments. 4. Chlamydia pneumoniae in a Free-Ranging Giant Barred Frog (Mixophyes iteratus) from Australia
The koala biovar of Chlamydia pneumoniae was identified in lung tissue from a sick, free-ranging giant barred frog (Mixophyes iteratus) by using electron microscopy, C. pneumoniae-specific fluorescent-antibody staining, cell culture, and sequencing of the ompA, ompB and 16S rRNA genes. This is the first report of a chlamydial strain infecting both a homeothe

5. Stable Haploid Cultured Cell Lines from frog Embryos*
Two haploid cell lines have been established from androgenetic embryos of the frog, Rana pipiens; one line has been maintained in culture for 150 generations, the other for 200 generations. Karyotypes of the two lines agree well with the standard for the species although some chromosomes show small differences in length. The cells multiply in the same define

6. The Presence of the Lucké Herpesvirus Genome in Induced Tadpole Tumors and Its Oncogenicity: Koch-Henle Postulates Fulfilled*
Herpesvirus extracted from a naturally occurring frog renal carcinoma (Lucké tumor) induced virus-free Lucké tumors in developing frogs. Herpesvirus recovered from an induced tumor after incubation at low temperature of tumor fragments cultured in vitro was oncogenic when injected into developing frog embryos. With the exception of the “pure culture” r

7. Elucidation of amidating reaction mechanism by frog amidating enzyme, peptidylglycine alpha-hydroxylating monooxygenase, expressed in insect cell culture.
A frog 'peptidylglycine alpha-amidating monooxygenase (PAM, EC 1.14.17.3)' was expressed in cultured insect cells by using the baculovirus expression vector system. The enzyme, recovered in the culture medium, was purified to homogeneity. Its apparent molecular mass (43 kd), estimated by both SDS-PAGE and molecular sieving, was higher than the value (39 kd)

8. Growth of Piscirickettsia salmonis to High Titers in Insect Tissue Culture Cells
Piscirickettsia salmonis was grown in established insect, frog, and fish tissue culture cells. The yield of P. salmonis in Sf21 cells was up to 100 times that obtained in CHSE-214 cells, and virulence for Atlantic salmon was retained. The ceiling temperature for growth of P. salmonis in Sf21 cells was 24°C.

9. Effect of Temperature on the Intracellular Growth of Histoplasma capsulatum
The rate and form of growth of Histoplasma capsulatum within histiocytes derived from homothermic and poikilothermic animals, and incubated at 25, 30, and 37 C, are described. The generation time of the fungus in mouse cells incubated at 37 and 25 C was 11 and 24 hr, respectively. Blastospore formation was progressively retarded in cells at 25 C, and this re

10. Acetylcholine release at identified nerve terminals in the organ-cultured frog neuromuscular preparation.
1. The frog cutaneous pectoris neuromuscular preparation was maintained in organ culture for a few days, at either 24 or 14 degrees C. The synaptic activity of identified nerve terminals was repeatedly examined in order to describe the sequence and time course of changes from normal synaptic activity to a complete synaptic silence. 2. We found that the 'tran

11. Replication fork barriers in the Xenopus rDNA.
To investigate replication fork progression along the tandemly repeated rRNA genes of Xenopus laevis and Xenopus borealis, rDNA replication intermediates from dividing tissue culture cells were analyzed by two-dimensional gel electrophoresis. Analysis of the direction of replication in the rRNA coding regions revealed replication forks moving in both directi

12. Stimulation of gastrin release by bombesin and canine gastrin-releasing peptides. Studies with isolated canine G cells in primary culture.
Bombesin, a polypeptide derived from frog skin, has been shown to stimulate gastrin release from the gastric antrum in vivo and in vitro. To elucidate the mechanisms of this effect, we developed a method to culture isolated and enriched G cells from canine stomach. After digestion of antral mucosa with collagenase and EDTA, dispersed cells were fractionated
