Sintese de macro esferas porosas de amino polimero : aplicação em imobilização de biocompostos

RESUMO

The work presented here concems the synthesis of N Methylolacrylamide (NMA) and Urea-formaldehyde (DF) resins through the polycondensation reactions of the acrylamide and urea with formaldehyde. Theses resins and the acrylamide monomers were used in the synthesis of macrospheres. Their solutions were dropped in hot silicon oil; after that macrospheres of poly (N-methylolacrylamide), poly (urea-acrylamide), polyacrylamide and polyacrylamide coated with N-Methylolacrylamide were obtained. The resins and the macrospheres were characterized by using infrared spectroscopy (FTIR). The macrospheres were analysed by scanning electron microscopy (SEM), mercury porosimetry and superficial area(BET). The macrospheres of poly (N-methylolacrilamide), poly (ureaformaldehyde) and polyacrilamide showed porosity as verified by electronic microscopy, mercury porosimetry and superficial area. The macrospheres were used in immunoassay, type ELISA(Enzime Linked Immunosorbent Assay), as support to immobilize mouse immunoglobulin(lg) in assays using mouse anti-immunoglobulin(a-Ig) labeled with horseradish peroxidase (HRP). The parameters as the pH of the imobilization, the concentration of Ig and a- Ig and the support were varied to analyse their influences in the ELISA assays. The polyacrylamide spheres were not analysed because they were soluble in the reaction medium, but the other macrospheres showed excell_nts results in the ELISA assays. The poly (N-metilolacrylamide) macrospheres showed the best results of all. Responses were obtained up to a diluition. In preliminary studies the macrospheres were used to immobilize lipase and then they were tested in the esterification reaction of the 3-Benzyloxy1,2-propenediol with vynil acetate. It was observed that the best pH value for the lipase immobilization is 4,0 but its catalytic efficiency was observed at pH=7,0
