Proteoglycans are important component of the extracellular matrix, which are involved in its organization and function, as well as in many biological processes. The aim of the present study was to investigate the changes that occur in proteoglycans of human fibroadenoma in comparison to proteoglycans of human normal breast tissue during the menstrual cycle. Tissue samples were collected during surgery, and both the concentration and the synthesis rate of the proteoglycans were analyzed. Fibroadenomas contained more, and synthesized more, proteoglycans than their paired normal controls, both in proliferative and secretory phases of the menstrual cycle. Normal breast tissue has shown an important increase in proteoglycan concentration during the secretory phase. In contrast, a small decrease was observed in the same period for tumors. Concerning the glycosaminoglycan chains, heparan sulfate and a hybrid dermatan sulfate containing both β-D-glucuronic and α-L-iduronic acid residues, were found in both normal and tumoral tissues. The concentration of both was increased in tumors, in comparison to normal tissue, but the dermatan sulfate increased more. The structure of this compound was also modified, with a higher proportion of β-D-glucuronic acid. Nevertheless, no differences were observed in dermatan sulfate composition comparing the proliferative to the secretory phase of the menstrual cycle. Decorin was identified in both normal and tumoral tissues, while versican was found only in fibroadenoma. By immunofluorescence, it was shown that decorin occurs mainly in peritubular regions. Versican, which again appeared only in fibroadenoma, also was concentrated in the peritubular regions. Our results clearly demonstrate that the synthesis rate and concentration of proteoglycans, especially decorin and versican, is increased in fibroadenoma in comparison to normal breast tissue. In normal breast tissue, the concentration of proteoglycans is higher during the secretory phase, but the tumor seems to be less sensitive to hormonal control.
