RESUMO

The small ruminantslentiviruses are responsible for a significant economic loss that enabled the development of efficient control and monitoring programs in Europe and other countries.The diagnosis of CAE is mainly based on serological tests.The World Organisation for Animal Health (OIE) recommends the use of agarose gel immunodiffusion (AGID) for its low cost, high specificity and practicality of implementation.The antigens used for serological diagnosis are composed mainly by structural proteins p28 and gp135.The AGID can determinefalse-negative results, dueto be a test of low sensitivity.Thus the production of viral antigens is important in testing for CAE, for this is a critical input that may influence the sensitivity of serological tests.In the protein extraction process, purification systems are used containing polymers or surfactants.The systems containing surfactants are simpler and less expensive.Currently, the SDS (sodium dodecyl sulfate) has been the surfactant preferably to extract and purifysmall ruminant lentivirus antigens.Therefore,this work aim to evaluate the performance of other surfactants in the caprine arthritis and encephalitis virus antigens production in relation to the surfactant SDS.The treatments evaluated by diagnosis method AGID was observed an improvement in the diffusion capacity of the antigens compared to the control not added with surfactants.The CHAPS and Triton X-100 surfactants had reactions to both: pellet and supernatant, indicating the presence of viral proteins, even with the presence of interfering as albumin in the supernatant.However, in Western blotting assays only the treatment with CHAPS showed a satisfactory result at all concentrations used.
