Produção, caracterização e aplicação de biopolímero de Agrobacterium radiobacter K84 como substituto de gordura

RESUMO
Agrobacterium radiobacter k84 biopolymer is a novel soluble metabolite obtained in this work by fermentation and proposed as a food ingredient. The process resulted in 10 g.L?¹ of a crude biopolymer composed of 40% minerals (P, K, Ca, Mg, Cu, Zn, Mn, Na, Fe, Cr, Co, Ni, Pb, Cd, Mo), 35% carbohydrates (glucose) and 15% proteins. In vitro biopolymer toxicity was evaluated using 2.5, 5.0 e 10.0 mg/mL in LLC MK2 (Rhesus monkey kidney) cell culture by the MTT test, resulting in cytotoxic concentration 50% (CC50) of 9.32 mg/mL. In a 30-day oral toxicity study, Swiss mice administered with biopolymer solutions by gavage at the doses of 25, 75, 250 e 750 mg/kg/day, presented no signs of harmful effects from hematological and histopathological analyses and the no observed adverse effect level (NOAEL) was 750 mg/kg/day. Concerning the technological properties, the biopolymer presented ability to form viscous solutions at room temperature, gelling activity, 83% water absorption, 69% solubility, 191% oil absorption and 56% emulsifying activity. As a partial and total fat substitute in chicken product (model system), the biopolymer had effect on texture being similar to the formulation with fat, but, as higher the fat amount in the formulation, higher the acceptation due to the taste, evaluated by hybrid hedonic scale test. Evaluation by light microscopy, showed emulsifying and stabilizing effects on the chicken product formulations with the biopolymer. Agrobacterium radiobacter k84 biopolymer composition, technological properties and absence of harmful effects make possible its applications in food, pharmaceutical and cosmetic products.
