Preparação e caracterização de lipossomas com a superficie modificada com gangliosidios para uso em imunoterapia

RESUMO

The pharmacological importance of gangliosides has increased in the last few years due to the identification of these compounds as antigens, producing antiganglioside antibodies, which may be involved in autoimmune diseases or neuropathy. Studies in ganglioside literature encompass their properties and functions in the neuronal membranes as well as their pharmacology in the central nervous system upon injection in pure ganglioside form in animais and humans. However, as associated to the liposomes, the studies of their effects on the immunological system are still scarce, which motivated the development of this research. This work includes the preparation and characterization of multilamellar liposomes consisting of dipalmitoyl phosphatidyl choline, cholesterol, dihexadecylphosphate and gangliosides, monosialoganglioside (GM1) and Mist (mixture of GM1 21%, disialoganglioside (GD1a) 40%, disialoganglioside (GD1b) 16%, trisialoganglioside (GT1b) 19%) and evaluation of immunologic effect of hese liposomes as administered in mice. The animais tested were Balb/c mice, which were treated with six liposome doses at 2 week-intervals during twelve weeks. After this period the animais were sacrificed and their sera were assayed by ELlSA tests in order to evaluate the production of antiganglioside antibodies of IgM and IgG classes. With respect to the statistical significance, the liposomes without gangliosides and the ones associated to the Mist were non immunogenic and some of animal sera, which were treated with liposomes associated to the GM1, showed some Immunoglobulin classes.
