RESUMO

Dextran is a polysacharide obtained fiom sucrose, a common and cheap raw material in Brazil. Its application depends on its molecular weight, that presents a large range. There are dextrans of molecular weight fiom thousands to millions of daltons. The enzymatic synthesis of dextran is possible because the enzyme used to obtain dextran is stable under large conditions, although the tradition process for dextran production is the fermentative one. The goal of this work is to study the viability of dextran enzymatic synthesis using a continuos system. The proposed system is based on the CARE process where the adsorption and dessorption of the enzyme are carried out in stirred tanks and the enzyme is recycled. In this way, the enzyme can be recovered and the production costs are reduced. The enzymatic reaction is carried out in the dessorption tank with the dessorbed enzyme. The CARE process was proposed protein purification. The system consists on two stirred tanks where the protein is adsorbed in the first tank and dessorbed and recoverd in the second tank. In this work a system is proposed which consists on three stirred tanks. In two of them the enzyme is adsorbed and in the another one the enzyme is dessorbed and the enzymatic reation takes place. The results obtained throughout the simulations using the three tanks system, proposed in this work, was compared to the ones obtained with a two tanks. A more realistic model, that takes in account the mass transfer resistances related to enzyme was also developed. The buffers flow rates were also considered in the system modeling. A hybrid model with neural networks was developed to predict the behavior of the two tanks system, in which the enzymatic reaction rate was obtained trough neural network. Based on experimental yields obtained under industrial conditions, the viability of the proposed system was verified for industrial applications
