RESUMO

Research for an American Tegumentary Leishmaniasis (ATL) test using venous blood, through Polymerase Chain Reaction (PCR), was performed in samples of two endemic counties in the State of Pernambuco, Brazil, in order to verify if the material was suitable for diagnosis. Venous blood from 119 subjects was collected: 63 from Cabo Santo Agostinho and 56 from Triunfo. Some tests were performed to stardardize the PCR and primers that amplified a 750 bp fragment of Leishmania Viannia braziliensis kinetoplast DNA (kDNA) were chosen because that species has been connected with ATL in Pernambuco. The results showed 48,7% of PCR-positive samples. From those, 31% belonged to individuals who had been treated and considered clinically cured (= no lesions). The findings show that: a) venous blood can be used for PCR in ATL diagnosis; b) the absence of lesions does not always correspond to a negative PCR; c) the presence of parasite DNA in treated patients calls attention to the possible presence of the very parasite and the consequent risk of ATL relapse; d)the treatment applied to the patients does not seem to be able to eliminate all the parasites
