Evaluation of the immune response to different antigens of Corynebacterium pseudotuberculosis in Canindé goats. / Avaliação da resposta imunológica de caprinos da raça Canindé a diferentes antígenos de Corynebacterium pseudotuberculosis.

RESUMO

Corynebacterium pseudotuberculosis is the etiologic agent of caseous lymphadenitis, a disease that affects goats and sheep, causing severe economic losses for creators in Brazil. In this study, four different antigenic extracts were obtained from the attenuated strain T1, which was isolated in Bahia State, Brazil. Forty-four Canindé breed goats were divided in five groups, receiving each one a different antigen solution and saline buffer as control. One of these antigens was the culture supernatant associated to Freunds incomplete adjuvant (FIA), two of them were this supernatant concentrated by three phase partitioning method (TPP) associated to an oligodeoxynucleiotide containing unmethylated CpG dinucleotides (CpG ODN) formulated with and without FIA. These groups received another immunization dose 28 days after the first one. Another group was immunized with alive strain T1 (2x105 CFU) this group does not receive a booster. Humoral response was monitored through the identification of specific IgG by indirect ELISA and Western Blotting, and the production of IFN-γ was followed, in order to observe the activation of cellular response. After twelve weeks of antigens inoculation, animals were challenged with 2x105 CFU of a wild strain, also isolated in Bahia State, and necropsy was performed in all animals twelve weeks after this challenge. It was observed that the immunization with whole attenuated bacteria gave a partial protection of 55.5%, besides the weak humoral response elicited. Animals inoculated with secreted antigen associated to Freunds incomplete adjuvant and CpG ODN showed a strong humoral response, but it cant prevent spread of challenge bacteria in the majority of animals. These results show the immunogenic potential of the attenuated strain T1 in the development of a vaccine against caseous lymphadenitis in goats.
