Estudo in vitro da atividade antimicrobiana imediata e mediata da clorexidina gel 2% usada como irrigante endodontico contra Enterococcus faecalis

RESUMO

The initiation and perpetuation of pulpal and periapical disease occur due to the presence of bacteria and their degradatory by products in the root canal. The host defense cells are unable to reach bacteria harbored in the root canal system thus, the root canal therapy must be performed combining mechanical procedures and chemical substances. For that reason, much attention has been focused on the chemical properties of the endodontic irrigants, seeking the infection control and the biocompatibility to the periapical tissues. Therefore, the aim of the present study was to assess in vitro the antibacterial activity of the 2% chlorhexidine gel against E. faecalis comparing to other endodontic irrigants (2% chlorhexidine liquid and 5.25% sodium hypochlorite). For this evaluation, 80 roots of human mandibular premolars were prepared by the serial instrumentation technique, autoclaved and contaminated for 7 days with E. faecalis monocultures. The roots were then divided into 5 groups according to the irrigant substance used during the standardized biomechanical preparation. In order to evaluate the antimicrobial action of the irrigants substances, 3 microbial samples were taken: pretreatment (before the biochemical preparation); post treatment (immediately after the biochemical preparation) and final (7 days after the biochemical preparation). The microbiological samples were plated to count the ColonyForming Units (CFU). Data were statistically analyzed by the program BioEstat 2.0. The 2% chlorhexidine gel and 2% chlorhexidine liquid reduced significantly the E. faecalis CFU in the post treatment and final microbiological samples. On the other hand, the 5.25% sodium hypochlorite also reduced the E. faecalis CFU in the post treatment however, bacterial growth were detected in the final sample. It was concluded that, among the substances assessed, the 2% chlorhexidine gluconate, gel and liquid, antimicrobial ability was more effective than the 5.25% NaOCI in keep low CFU of E. faecalis atter 7 days
