RESUMO

Eighty-three specimens of propolis which were collected by africanized Apis mellifera in various regions of Brazil were extracted by 80% etanol and examined for concentrations of total flavonoids in ethanol extract of propolis by methods of both chemical analysis, and Ultraviolet-spectrophotometer (U.V scanning). The results of concentrations of total flavonoids in propolis which were determined by chemical analysis correlated with the results obtained by U. V. scanning. The U. V. scanning showed a maximum absorption spectra of flavonoids at 290 nm, The qualitative analysis of flavonoids in propolis was determined by High Performance Thin Layer Chromatography (HPTLC) and Reversed phase High Performance Liquid Chromatography (HPLC). The results of HPTLC analysis of ethanol extracts of propolis demonstrated patterns of fractions of flavonoids when irradiated by U.V. at 254 and 366 nm, respectively, but at 366 nm showed more distincts patterns of flavonoids. These results demonstrated different patterns among propolis from the States of São Paulo (SP), Minas Gerais (MG), Goiás (GO), Mato Grosso do Sul (MS) as compared to those from Paraná (PR) and Rio Grande do Sul (RS). Quantitative concentration of flavonoid fractions were determined by HPLC. The profiles of HPLC also demonstrated different patterns among samples from SP, MG, GO, MS and PR, RS. Furthermore, propolis from RS and PR contained more quercetin, galangin, chrysin as compared to those from SP, MG, GO, MS, but propolis from RS and PR contained less acacetin and no isorhamnetin. These results demonstrated that the concentrations of total flavonoids were variable depending on the geographic location and plant ecology.
