Estudo cinetico e de equilibrio na separação cromatografica dos enantiomeros da cetamina em triacetato de celulose microcristalina

RESUMO

There is a growing demand for efficient and low cost methods for the purification of the stereoisomers from chiral molecules, named enantiomers, in the manufacturing of drugs, flavours, pesticides and food additives, where these compounds are commonly encountered. Chrornatographic method is considered in nowadays an important alternative for the production of enantiomers with high purity, especially for compounds applied to biological systems, which discriminate between the two isomers. The development of preparative processes, the experimental study and the modelling of columns where the chromatographic processes of adsorption and elution take place depend strongly on the experimental determination of some fundamental parameters. These are the total and intersticial porosity of the bed of adsorbent particles, and equilibrium and mass transfer parameters. Columns employed in the separation of racemic mixtures are packed with stationary phases constituted of micrometric porous partic1es. The development of separation processes at preparative scale are accomplished based on measurements made with small size columns packed with the same stationary phase. A systematic procedure for the determination of parameters was developed in the present work through the method of moments with the utilization of tracers with different molecular weight and adsorption capacities, allowing the measurement of the total porosity of the bed and the porosity of the partic1es of the stationary phase microcrystalline cellulose triacetate. The coefficients of axial dispersion and mass transfer were determined through injections of racemic ketamine, for five different temperatures ranging from 25 to 50°C. The adsorption isotherm was determined in the temperature of 25°C, and the effect of the amount of injected solute on the peak shape was studied. The results of this work will represent a base for the project of the experimental conditions of a preparative simulated moving bed plant
