RESUMO

A carbon paste electrode modified with enzyme laccase productNovozym 51.003 has been proposed in this manuscript. This enzyme catalyzesthe oxidation of different types of phenols compounds, and lignin aminefunctional groups engaged. This enzyme has been among the most applied inthe construction of biosensor amperometric Electrical. The effect of laccaseamounts added on the carbon paste was studied using 50ìL, 100 ìL, 150 ìL e200 ìL of the product Novozym 51.003 i. The mixture of carbon paste wascomposed by 75% (m/m) of the graphite and 25% (m/m) nujol. The mediatorABTS (salt diamine of 2,2- azino- bis (3-ethylbenthiazoline-6-sulfonic acid)influence and pH were also evaluated. As support to immobilize modifyingagent was used syringes plastic 1 mL (syringes Nicas to insulin), adapted with acopper wire attached to the piston connected to the contact externally withgraphite rod. Another technique use to immobilized enzyme was Layer byLayer. In this step was used, FTO glasses (tin oxide doped with Fluor, Flexitec)as support. The materials used were laccase enzyme (Novozym 51003);polyeletrolytes PDDA (poly (diallyldimethylammonium chloride) and differentsources of starch: corn and cassava. Parameters such as pH and scan ratewere studied at this stage and demonstrated that for the electrode constructionof the immobilized enzyme according to the setPDDA/CASSAVASTARCH/PDDA/LACCASE was the most higher responseand the better electrode stability.
