RESUMO

Objective: To analyze the effects of metformin on the reproductive system of neonatally androgenized female rats that developed permanent estrus. Materials and methods: A total of 75 three-day-old female albino wistar EPM-1 rats were randomized to three 25-animal groups: control (CG), androgenized (AG), and androgenized + metformin (AGmet). The animals in AG and AGmet were administered 0.1 mL of testosterone propionate (1.25 mg/animal) diluted in castor oil (vehicle), and controls were given vehicle only. The injection site was in the dorsal subcutaneous region. Nine animals died during drug administration. After 90 days a daily six-week gavage treatment was initiated with the rats in CG and AG receiving vehicle (distilled water) and those in AGmet getting metformin (50mg/kg). Afterwards, 12 animals from CG, 10 from AG, and 14 from AGmet were sacrificed. Blood was collected for biochemical measurements and the uteri and ovaries were extracted for histological analyses (experiment 1). The remaining 30 animals were mated (experiment II). Data analyses were carried out with Tukeys test and ANOVA. Results: In terms of fasting glucose and HOMA-IR, indices were highest in AG (p<0.01) and there was no significant difference between AGmet and CG. Histomorphometric analyses of the ovaries and uteri revealed, in AGmet, reduction in the area covered by degenerating follicles, reduction in the number of interstitial cells and identified corpora lutea , diminution of endometrial thickness, and a decrease in the amount of collagen and the number of eosinophiles, typical in comparison with AG (p<0.01). After mating, spermatozoids were identified in the vaginal ring, in all of the animals in CG (n=10), four in AGmet, and none in AG. All of the animals in CG had full-term deliveries, but only one in AGmet. Conclusion: Metformin led to improvement of the fasting glucose and HOMA-IR index and to partial reversion of the histomorphometric characteristics of the ovaries and the uterus in neonatally androgenized female rats.
