RESUMO
Several studies have been developed in the intention of already improving the protocols existent for the freezing of canine semen. With that the present work had two objectives: 1) to evaluate the effect of the addition of different dimethylformamide concentrations on the characteristics of the canine semen diluted in coconut water in the powder form (ACP-106C) and frozen, 2) to Compare the effect of the temperature of addition of the dimethylformamide (27 and 4C) on the canine semen diluted in ACP-106 C and frozen. Ten ejaculated originating from of five dogs were collected by digital manipulation. For the first experiment, all the aliquots were diluted in ACP-106C containing 10% of egg yolk, and in the first aliquot it was used as crioprotetor the glicerol 6% (I group control-GC), Monday 2% of dimethylformamide (DF2), the third 4% of dimethylformamide a (DF4) and Wednesday 6% of dimethylformamide (DF6). Already in the second experiment, a semen sample was diluted in ACP-106 C with 10% of egg yolk and glicerol to 27C (G27), Monday diluted in ACP-106 C with 10% of egg yolk and dimethylformamide to 27C (DF27) and the third diluted in ACP-106 C with 10% of egg yolk and dimethylformamide to 4C (DF4). The samples of the two experiments were submitted to the freezing process, being conditioned soon after in liquid nitrogen. After one week, the semen samples were thawed her/it a temperature of 37 C for 1 minute, being then submitted to the morphology evaluations, integrity acrossomal, test hiposmótico, as well as the computerized analysis. In the first experiment, the total mobility of GC was superior to the one of the groups containing dimethylformamide. The population of spermatozoids fast, slow and static was significantly larger in GC compared to the other groups. In the second experiment, a superiority of the group was evidenced it controls (G27) in relation to the groups containing dimethylformamide for the parameters progressive mobility, VAP, VCL, VSL, LIN, STR and ALH. Before the obtained results it is ended that the dimethylformamide in the concentrations added to the diluidor ACP-106C and 10% of yolk, it provides resulted in vitro considered insatifastórios for the for freezing of the canine semen, not being indicated your use. Besides, there is not influence of the temperature of addition of the dimethylformamide the use of the diluidor ACP-106 C, in the quality of the canine semen.
