Determinação da microbiota bacteriana de mosto e de dornas na fermentação alcoolica

RESUMO

Samples of concentrated yeast cell suspension diluted before the addition of sulfuric acid, of the same concentrated yeast cell suspension after addition and action of sulfuric acid, of must added to the same concentrated yeast cell suspension collected and of fermented must from the same fermentation tank, were collected between July and December of 1986, at the Costa Pinto Sugar Mill, Piracicaba - ap, to determine the contaminant bacterial micro flora from alcoholic fermentation. Aerobic plate counts were made in PCA and MRSA added of 10 ppm of cicloheximide (actidione), and 353 colonies were selected, of which 19 were lost and 334 were utilized at the present work to be classified by the use of probability matrices. It was found that Gram-positive (98,52%), ( 73,95%) . Lactobacillua sp. were the major contaminant (59,75%), followed by strains of the Bacillus (26,58%), Staphylococcua (8,76% ) , Micrococcus (1,56%), strains of the Enterobacteriaceae Family (1,48%), Pediococcua (1,26%) and Streptococcus (0,70%). The Lactobacillus strainswere classified as L. fermentum (15,04%), L. helveticus (14,08%), L. plantarum (5,69%), L. animalis (4,55%), L. buchneri (3,76%), L. acidophilus (3,07%), L. vitulinus (2,96%), L. viridescens (2,35%), L. amylophilus (1,88%), L. agilis (1,25%), L. reuteri (1,22%), L. delbrueckii subsp. lactis (1,04%), L. murinus (1,02%), L. delbrueckii subsp. bulgaricus (0, 71%), L. coryniformis subsp. torquens (0,71%) and L. sake (0,42%). The Bacillus strains were classified as B. coagulans (15, 09%) , B. stearothermophilus (6,91 %), B. megaterium (2,43%), B. brevis (1,23%), B. lentus (0,70%) and B. pasteurii (0,22%). The Staphylococcus strains were classified as S. cohnii subsp 1 (3,10%), S. caseolyticus (1,93%), S. cohnii subep. 2 (1,42%), S. hyicus subsp. hyicus (0,84%), S. auricullis (0,79%) and S. epidermidis (0,68%). Micrococcus roseus (1.56%) was the only specie of Micrcocus. The Pediococcus strains were classified as P. acidilactici (0.87%) and P. inopinatus (0.39%). Streptococcus pneumoniae (0.70%) was the only specie of Streptococcus. It was found that the acid treatment with sulfuric acid, by more or less two hours at ph 2.5, decreased in 44.55% the contaminant bacterial micro flora initially present.
