RESUMO

The aim of this research was to study the embryonic development at different stages, the influence of diverse concentrations of MT medium (Murashige &Tucker, 1969), sucrose, vitamins, activated charcoal and gibberellic acid on the culture of immature embryos from cross between âPÃra Rioâ x âPoncÃâ. The embryos were excised under aseptic conditions and inoculated in 15 mL of MT according to each of the following experiments: 1) stages of development (globular, heart-shaped and cotiledonary) associated with concentrations of 0%, 50%, 100% and 150% of the MT medium; 2) MT concentrations (0%, 50%, 100%, 150% and 200%) combined with 0, 30, 60 and 90 g.L-1 of sucrose; 3) vitamins concentrations of the MT (0%, 50%, 100%, 150% and 200%) combined with 0, 30, 60 and 90 g.L-1 of sucrose; 4) activated charcoal concentrations (0, 0.5, 1, 1.5 and 2 g.L-1) combined with GA3 (0, 0.01, 0.1; 1 and 10 mg.L-1). After the inoculation, the embryos were maintained for 90 days in growth room at 27Â1ÂC temperature, 16-hour photoperiod and 32 umol.m-2.s-1 of irradiance. High development was obtained with embryos at cotiledonary stage. High development at the globular embryos stage were obtained using 50% of the MT added to 60 g.L-1 of sucrose and 100% of the same medium added to 90 g.L-1 of sucrose. Both supplemented with 0,01 mg.L-1 of GA3. There was no need to add activated charcoal and vitamins in the MT to globular embryos culture.
