RESUMO

The control and treatment of dogs with synthetic drugs have not shown effectiveness in reducing canine visceral leishmaniasis. In an attempt to find new agents with leishmanicidal action, plant compounds have been investigated. The objective was to determine the in vitro activity of fractions of Musa paradisiaca (banana tree) and Spondias mombin (cajazeira) of Leishmania chagasi. The fractions were obtained by column chromatography of crude methanol extracts of plants. In vitro tests were performed on promastigotes and amastigotes of L. chagasi MHOM46/LC/HZ1 and included the cytotoxic effect of these fractions in RAW 264.7 cells. The strains of L. chagasi were grown in medium M199 plus 10% fetal calf serum, 5% sterile human urine and gentamicin (40mg/ml) in a chamber at 23.6 C. The fractions were diluted and added to promastigotes at concentrations of 100, 50, 25, 12.5 and 6.25 μg / ml overnight. The viability of parasites was evaluated using the tetrazoliumdye (MTT) colorimetric method. Subsequently SDS 10% and 4N HCl were added and read at 570nm using a microplate reader. The experiment had as positive control pentamidine (40μg/ml) and as negative control the medium without treatment. The amastigotes were subjected to in situ ELISA and the reading performed at 492nm. Had as positive control amphotericin B (40 g / ml) and as negative control medium with cells and promastigotes without treatment. The fractions that have the best results on promastigotes were S.mombin - 100% methanol and acetate: methanol (80:20) with IC50 (inhibitory concentration 50% of parasites) of 58.1 μg / ml and 11.26 μg / ml, respectively. The fractions of S. mombin acetate: methanol (70:30), acetate: methanol (80:20), acetate, chloroform and the fraction (90:10) hexane: chloroform (50:50) from M. paradisiaca, had better action on amastigotes in the concentrations: 10.68 μg / ml, 0.2734 μg / ml, 0.6141 μg / ml and 15.07 μg / ml. All results obtained on the promastigotes were statistically similar to the positive control pentamidine whose IC50 ranged from 0.01 to 497.3 mg / ml. All values obtained for IC50 on amastigotes were statistically similar to those obtained by standard drug amphotericin B ranging from 0.58 to 28.68 μg / ml. These natural substances were not cytotoxic to RAW 264.7 cells. These results indicate that these compounds have inhibitory activity in vitro on promastigotes and amastigotes of L. chagasi and suggest future studies to evaluate the effects in vivo.
