Análise comparativa das variações morfológicas entre dois diferentes isolados (Pb01 e Pb18) do fungo Paracoccidioides brasiliensis e da sua interação com o sistema imune do hospedeiro

RESUMO

The dimorphic fungus Paracoccididoides brasiliensis is the etiologic agent of systemic mycosis paracoccidioidomycosis, it is restrict to Latin American. It is frequently diagnosed in Brazil, Colombia and Venezuela. The evolution of this disease depends directly on the interaction between P.brasiliensis and host. Aspects of funguss virulence and the immune type response of host that is caused by fungi are important to understand differences on the clinical forms disease. An important aspect to be considerated on this interaction are genetic differences, observed between phylonegetics groups from Paracoccidioides genus. In this work we analyzed morphological differences of two isolate of P. brasiliensis (Pb01 and Pb18) and studied the interaction of these isolate and macrophages in vitro. Immune response induced by distinct ways of infection in murine was caracterizated. The isolate Pb01 cells showed mainly a circular shape and a littlest diameter either to mothers cells and budding compared to Pb18 that presented an elliptical and circular shape. Yeasts Pb01 are phagocyted fastly and in a major quantity than Pb18. Yeasts Pb18 survived for more time inside macrophages and induced a little oxide nitrics production. In systemic infection, inoculated animals with Pb01 presented more efficient linfoproliferative response until 30 days after infection. After this period, the response became similar to Pb18, which causes suppression of linfoproliferative response. In this kind of infection we observed an immunological response predominantly Th1. But, in infection by intratracheally way occurred a mixed immunological response (Th1 e Th2) in infected animals, however in two types of infection, a low fungics load was recovered from lungs tissue and did not present formation of inflammatory granulomatosa response. Infected animals with isolate Pb18, in both ways of infection, occurred mainly response Th2 and a crescent increase on quantity of fungal burden recovered from lungs and an inflammatory granulomatosa in this organ. Then, the isolate Pb01 has a minor pathogenicity in relation to Pb18 and induces an immunological response more efficient in model animals.
