1. Diagnosing lysosomal storage diseases in a Brazilian non-newborn population by tandem mass spectrometry
OBJECTIVES: High-throughput mass spectrometry methods have been developed to screen newborns for lysosomal storage disorders, allowing the implementation of newborn screening pilot studies in North America and Europe. It is currently feasible to diagnose Pompe, Fabry, Gaucher, Krabbe, and Niemann-Pick A/B diseases, as well as mucopolysaccharidosis I, by tan

Investigation of Carboxypeptidase, Sphingomyelinase and Alkaline Phosphatase from Schistosoma mansoni as potential vaccine candidates.

4. Effect of dimethylsufoxide on sphingomyelinase activity and cholesterol metabolism in Niemann-Pick type C fibroblasts
Niemann-Pick type C (NPC) fibroblasts present a large concentration of cholesterol in their cytoplasm due to a still unidentified deficiency in cholesterol metabolism. The influence of dimethylsulfoxide (DMSO) on the amount of intracellular cholesterol was measured in 8 cultures of normal fibroblasts and in 7 fibroblast cultures from NPC patients. DMSO was a

In the present study the recombinant form (rLiD1) of a dermonecrotic protein from the Brazilian Loxosceles intermedia spider venom was expressed in Escherichia coli and purified by reversed-phase HPLC. Twenty five mg of rLiD1 was produced from liter of bacterial culture. The molecular weight of rLiD1 was verified by mass spectrometry (32,758 Da). rLiD1 displ

Participação das plaquetas na dermonecrose induzida experimentalmente pelo veneno loxoscélico em coelhos / Contribution to the study of the in vitro action induced by the venom of the brown spider, Loxosceles gaucho, on human and rabbit platelets. Platelet participation in the dermonecrotic lesion experimentally induced in rabbits by the loxoscelic venom
Venoms from Loxosceles spiders exhibit a wide range of activities on different cells, tissues and systems, being specially evident their ability to cause a dermonecrotic lesion. In a previous paper, we showed that Loxosceles gaucho spider venom induces intense thrombocytopenia in rabbits. Thus, in the present study, we firstly aimed to evaluate the in vitro

8. Protein-mediated surface structuring in biomembranes
The lipids and proteins of biomembranes exhibit highly dissimilar conformations, geometrical shapes, amphipathicity, and thermodynamic properties which constrain their two-dimensional molecular packing, electrostatics, and interaction preferences. This causes inevitable development of large local tensions that frequently relax into phase or compositional imm

9. Caracterização imunoquímica e molecular da fração dermonecrótica do veneno da aranha marrom Loxosceles intermedia
To investigate the biochemistry and the immunochemistry of proteins from L. intermedia spider venom, thirteen monoclonal antibodies were produced. On of them, called LimAb7, was able to neutralize the dermonecrotic activity induced by venom in rabbits. In an effort to characterize the proteins recognized by the neutralizing monoclonal antibody, we have used d

10. A new form of Niemann-Pick disease characterised by temperature-labile sphingomyelinase.
A new type (F) of Niemann-Pick disease characterised by childhood onset of splenomegaly, lack of neurological involvement, and diminished sphingomyelinase activity is described. The clinical presentation and heat-labile sphingomyelinase activity of this type F Niemann-Pick disease distinguishes it from other types of Niemann-Pick disease.

11. Copurification of Leptospira interrogans serovar pomona hemolysin and sphingomyelinase C.
The hemolytic and sphingomyelinase C activities of supernatants of cultures of Leptospira interrogans serovar pomona tended to copurify when isoelectric fractionation was carried out. Both activities focused primarily at pH 8.1. Considered in conjunction with other circumstantial evidence, the results led to the conclusion that sphingomyelinase C is responsi

12. Apolipoprotein C-III-1 activates lysosomal sphingomyelinase in vitro.
Apolipoprotein (apo)C-III-1 from human very low density lipoprotein stimulates 14-fold the activity of lysosomal sphingomyelinase from human fibroblasts. At the sphingomyelin concentrations tested, maximal stimulation was obtained with 5 microM apoC-III-1 or apoC mixture. Apolipoproteins A-I, A-II, B, and C-I conferred little or no stimulation.
