RESUMO

The objetive of this study was to evaluate the dimethyl formamide and its association effects on the goat semen cryopreservation, through the semen physical analyses and complementary tests, carried out during the cryopreservation process. Alpine and Saanen bucks from the Federal University of Viçosa Dairy Goat Experimental Section- DZO were used. The semen samplings were frozen with the yolk-skimmed milk diluent, associated with different cryoprotectors and concentration: glycerol 7%; (T1); glycerol 3.5% with dimethyl formamide 3.5% (T2) 5% dimethyl formamide5% (T3). The semen were centrifuged, diluted and bottled in a 0.5 mL straws. These straws were cooled at 5.0C for more than 1:20h. Then they were submitted to liquid nitrogen vapor for 15 minutes and immersed in N2 liquid. The thawing was made in water bath at 37C for 50 seconds. The parameters evaluated in vitro were progressive motility and sperm vigour, acrossome integrity, plasmatic membrane integrity (HOST) and acrossome reaction. A 30.0% initial motility loss was observed when the samples were submitted to the cryopreservation procedures. There was a 19% plasmatic membrane integrity loss, evaluated through the hipoosmotic swelling test (HOST). The acrossome lesions increased 3% during slow TTR (P<0.05). No differences were observed in the motility aspects among the sperm from the treatments, motility aspects at 0.00h; 0.05h; 1.00h of TTR and vigor in all the times given (P<0.05). At 2.00h of TTR a difference between the treatments (1) and (2) was observed (P<0.05), with the T2 semen motility superior to the others (P<0.05). The plasmatic membrane integrity values, acrossome integrity and acrossome reaction after the thawing did not differ between the semen from both treatments (P>0.05). These results show that the dimethyl formamide can be an alternative in the goat semen cryopreservation, when used alone or associated with glycerol.
