RESUMO

Carrot tissue were obtained from the secondary phloem region of adult roots from eight cultivars in order to test the tolerance/susceptibility to high levels of manganese. The cultivars studied showed differences in requirements of growth regulator for induction of callus. Liquid cultures were obtained from an initial inoculum of 104 - 105 cellsml. The maximum concentration of cells in suspension was 1.25 x 105 cellsml, with renewal of 30% of culture medium at 2 - 3 days interval. The MnC12 as source of Mn allowed higher fresh weight of callus when the concentration of manganese was between 0.1 to 32.0 mM. The MnSO4 as source of Mn gave higher concentration of endogenous manganse in the tissue. No differences in growth were observed due to the initial pH. The addition of 1.0 to 60.0 uM of IAA did not change callus growth when concentration of manganese was changed. The reduction in the concentration of phosporus, Calcium and magnesium in the initial culture medium provided higher availability of mangenese. With this modified medium, the range in manganese concentration in which the tissue of the cultivars studies showed variation in tolerance susceptibility, was from 0.025 to 8.0 mM.
