RESUMO

We evaluated the potential of muscle autografts to perform primary nerve repair when used immediately after their preparation with lidocaine injection. The right sciatic nerve of adult Wistar rats was sectioned and muscle grafts were used 15 minutes or 24 hours after lidocaine injection, to repair a gap 1.0 cm longo Morphometric analyses showed a decrease in axon and nerve fiber diameter, as well as in myelin thickness, which were similar for both types of grafts and significantly different ftom the control. G-ratio values in the distal stump for both groups were not different ITom the normal sciatic nerve, showing that a proper leveI of mye1ination was attained by the regenerated axons. However, within the graft, the G ratio was increased in both groups, suggesting poorer myelination in the graft region. Ultrastructural profiles of intramuscular nerve bundles and neuromuscular junctions, for both 15 minutes and 24 hours after graft implantation were similar to the normal. These results suggest that there are no restrictions to the use of a muscle graft immediately after lidocaine injection, in primary nerve repair, since the results of nerve regeneration were comparable to those observed when this type of graft is used 24 hours after being prepared. The decrease in myelin thickness in the graft suggests that the associations between Schwann cells, regenerated axons and muscle basal lamina should be further investigated to improve muscle graft nerve regeneration
