RESUMO
The aim of the present investigation was to study the efficacy ofaxonal regeneration after sciatic nerve cut and repair by muscle basal lamina autografts. It was done in comparison with axonal regeneration through nerve autograft. In one group of animals, muscle basal lamina autograft was obtained by injecting soleus muscle of adult ice with 2% lidocaine hydrochloride. Twenty four hours later a fragment of the injected muscle was dissected out and trimmed in order to get a graft big enough to fil! a gap of 5 mm. The left sciatic nerve was then cut and repaired with the graft held by epineural sutures at the proximal and distal nerve stumps. In another group, a fragment of 5 mm was dissected out from the left sciatic nerve and replaced at the same site by epineural sutures. Fifty days after surgery, animals from both groups (nerve autograft and muscle basal lamina autograft) were sacrificed and three fragments of the left sciatic nerve (distal and proximal to the grafted site and the grafted site itself) were processed for electron-microscopy. Ultrastructural analysis showed that there were no differences between regenerated axons in muscle autograft and nerve autograft. Also the overall arrangement of the axons at the graft site was in accordance with that described in the literature. Quantitative results show that in both cases axons crossed the graft site and reached distal stump. We noticed that there are no significant differences on the axonal regeneration between nerve autograft and muscle basal lamina autograft. On the nerve autograft we observed 13.592 `+ ou -´ 1420) myelinated regenerated axons on the distal stump, and on the muscle basal lamina autograft 13.792 `+ ou -´ 1145). These differences were not statistically significant. We conclude that muscle basal lamina grafts offer conditions that allow axonal regeneration in a very similar way to that observed with a regular nerve autograft
