Quantitative and qualitative analysis of in situ bacterial plaque retention in composites with smooth and rough surfaces / Analise quantitativa e qualitativa do acumulo de placa bacteriana in situ em resinas compostas com superficies lisas e rugosas

RESUMO

The aim of this study was to evaluate the effect of surface roughness of different filled composites on in situ bacterial plaque retention, quantified by spectrophotometry and qualified for mutans streptococci group by polymerase chain reaction (PCR). Specimens were prepared with the composites Durafill VS, Tetric Ceram and Filtek P60, with smooth and rough surfaces, produced by phtotoactivation through Mylar strip or Sof-Lex discs, respectively. The interaction composite x texture surface generated six experimental groups: Durafill VS/smooth surface, Durafill VS/rough surface, Tetric Ceram/smooth surface, Tetric Ceram/rough surface, Filtek P60/smooth surface and Filtek P60/rough surface. Surface roughness of samples was evaluated by profilometer reading (Ra). For in situ experiment, ten subjects were chosen which wore acrylic palatal appliances containing samples of the six experimental groups. All subjects wore the appliances for three days a week. In the fourth day of the first six weeks, the amount of dental plaque formed on the samples were extracted in 1.0M NaOH and analyzed by spectrophotometry. Rough surface samples showed significantly greater amount of bacterial plaque than smooth surface samples, for the three composites tested. For the smooth samples, in spite of the significant difference between the lower roughness of Durafill VS composite and the higher roughness of both Filtek P60 and Tetric Ceram composites, no difference was observed in the amount of bacterial plaque among the tested materials. In the seventh week, DNA of the bacterial plaque formed on the samples was extracted and compared by PCR using universal primers for mutans streptococci group, which amplify ribossomal RNA 16S genes, and digestion of PCR products with the restriction enzymes HaeIII, CfoI e RsaI. Bacteria of mutans streptococci group were found in all composites samples. The profiles generated by the PCR-RFLP analysis of the volunteers samples were similar, indicating that the composites Tetric Ceram, Durafill VS or Filtek P60 and the smooth or rough surfaces did not affect significantly the composition of the bacterial plate formed on the samples for mutans streptococci group
