RESUMO

Lipases are most commonly found in nature, they can be obtained from animals, vegetables and microorganisms and it can be used on different industries. The industrial lipase production increased considerably in past few years, such as they have innumerous application on different industrial processing. Nowadays, lipases studies analyzed some steps of production as up stream and down stream steps, enzymatic production, scale up and industrial application. These factors together determine the viability of industrial process for enzymatic production. This work analyzed the steps of up stream (previously clarification of fermentation medium and the optimization of inoculum conditions), fermentation (optimization of fermentation medium composite), scale up (enzyme production in stirred and airlift reactors), down stream (lipase purification for hydrophobic interaction chromatography) and partial characterization of lipases obtaining from Geotrichum candidum NRRL Y-552 This work showed that is viability to produce lipase changing conventional nutrients (as peptone and yeast extract) for industrial residues as nitrogen sources. Three fermentation media was optimized to lipase production (medium 1: 3.5% of yeast hydrolisate and 0.7% of soy oil; medium 2: 8.0% of corn steep liquor and 0.6% of soy oil and medium 3: 12.0% of clarification corn steep liquor and 0.6% of soy oil). The maximum lipase activities were of 20.0 U/mL in shake flasks and 24.0 U/mL in airlift reactor, values so high that those was reported in literature of the same microorganism. Then, the set of studied of previous clarification and enzymatic recovery allowed to associate the viable of purification process with cost reduction obtained using industrial residues as nitrogen source. In addition, highest level of lipase activity was possible using a no conventional reactor (airlift), due this reactor produced slowly damaged in cell mycelium. All these factors associated indicate that it is very promising the possibilities of utilization of this process on industry.
