MicropropagaÃÃo calogÃnese e irradiaÃÃo da figueira "Roxo de valinhos" / Micropropagation, callogenesis and irradiation of fig plants âRoxo de Valinhosâ

RESUMO

It was carried out three in vitro works with âRoxo de Valinhosâ fig plants. The first one aimed to verify the sensibility of explants to gamma radiation and the plantlets were separated by size (2,5 to 4,5; 5 to 7 and 8 to 10 cm) and were taken to the Nuclear Energy Center, irradiated on the doses 10, 20,30,40 e 50 Gy and cutted separating the bud portion: apical, medium and base. It was evaluated 90 days after inoculation by mortality, anomalies, number of leaves and length of aerial part. On the second work, it was evaluated the efficiency of nodal and leaves explants, the effects of light and dark and the best combination of 2,4D and kinetin at 0; 0,5; 1,0; 2,0 e 4,0 mgL-1 in the in vitro callogenesis. The characteristics evaluated were: growing intensity and weight of fresh mass of calluses. The third work aimed to optimize the micropropagation protocol testing: 1) alteration on WPM concentration - 0, 50,100,150 e 200% and the sucrose â 0,10,20 e 40g; 2) inoculation of nodal explants with 1,2,3 and 4 buds in combination with different doses of kinetin at 0; 0,5;1,0; 2,0 e 4,0 mgL-1.The evaluation were carried out 90 days after the inoculation by: root and aerial part high; weight of root and aerial part. The results revealed that doses until 50 Gy do not cause death of plantlets; doses over 30Gy block root formation, plantlets size 2,5 to 5 cm irradiated at 10 Gy present higher lengths of aerial part and number of buds and the increase of irradiated doses caused reduction on the weight of plantlets in any size of plantlets. The callogenesis on fig plants âRoxo de Valinhosâ can be obtained by leaves explants, absence of light and presence of 2,4D and kinetin at 4 mgL-1. On the sprouts multiplication can be taken 100% of WPM medium added with 10gL-1 of sucrose. When it is wished elongated sprout the use of medium without kinetin and explants with 1 or 2 buds promote best results. Adding 0,5 mgL-1 of kinetin and taking explants with 3 buds promote larger sprouts number.
