Metabolismo de radicais livres durante o ciclo estral de ratas

RESUMO

The hormones variations during the estral cycle bring morphologic and functional variations to different involved organs in the reproductive process, among them ovary, adrenal and uterus. The hormone variation as the E2 and the LH, that exert influence on the metabolism of the free radicals, also demonstrates that to these chemical species participation of the alterations of the reproductive cycle. Using rats as experimental models for the study of the reproductive cycle, daily vaginal smears was become fullfilled to determine its regularity and the phase of the cycle in each animal if it found, being sacrificed after 21 days, taking the ovary, uterus and adrenal, after the samples were divided in four groups according to the phase where the animals at the moment of the sacrifice met. The oxidative damage in uterus was quantified by the TBARs method and xylenol orange, carbonyl protein and the levels of the endogenous glutatione. Three enzymes of antioxidant system had activities determined by GR, GPx and GST in the three different tissues, adrenal, ovary and uterus. Our results demonstrate that to have an increase of oxidative damage in lipids and proteins in Diestro II, followed for an increase of the levels of GSHeq and the activity of the GST. In the ovary it exists together with an increase of the levels of lipid peroxidation in the Estrus and an increase of the oxidativo damage in proteins in diestro II with a reduction in the GSHeq/GSSG relation and an increase of the activity of GPx. In the adrenal changes throughout the cycle in the levels of lipid peroxidation and proteins it was observed a reduction in the relation of GSHeq/GSSG met in the phase of diestro I. The numbers show the presence of oxidative stress in the uterus in the second half of the reproductive cycle, which seems to provoke in the organ a reply with the increase in the activity of GST and the GSHeq levels, this oxidative stress could be related with the inflammatory alterations that occurs in the endometrium at this moment of the estrous cycle. In the ovary the lipid peroxidation in estrus shows an oxidative damage that can occur in membranes that could lead to a follicular rupture, and the presence of oxidative stress in the diestro I and II represented for the carbonyl protein results and the levels of glutationa could be associated to the phenomenon of luteolise. In the adrenal occurs a oxidative stress in diestro I. Thus an increase of oxidative stress in the ovary, uterus and adrenal seems to have a physiological paper next to these organs.
