Influencia do uso topico episcleral intra-operatorio de mitomicina C na proliferação e diferenciação de celulas epiteliais da cornea de coelhas

RESUMO

This study was performed to evaluate the influence of 0,02% mitomycin C (MMC), applied on a single dose for 3 minutes, in proliferation and differentiation of the cornea rabbits epithelium cells. During the surgery procedure, MMC or 0,9% physiologic saline solution (SF) was applied and the solution for each animal (both eyes) was performed by raffle. A topic solution was applied at episclera of the temporal limbic area, with intact epithelial tissue (one eye) and after partial corneal desepithelization (other eye). The animals were arranged in a group A (20 eyes), group B (16 eyes) and group C (14 eyes). They were sacrificed respectively at 4th , 15th and 45th days of the post-operative. To stimulate cell proliferation, the animals of group C were submitted to central corneal desepithelization 3 days before the sacrifice day. Cellular differentiation markers (AE1, AE3 and AE5) were used for differentiation study of the corneal epithelium. A cellular proliferation marker 5-Bromo-2-Deoxyuridine (BrdU) was applied by intravenous injection in all of the animals, 24hs before the sacrifice. In the slides labeled with BrdU technique, the nasal, temporal and central areas were determined under optic microscopy. The number of labeled cells was counted in 3 different fields of each area using millimeter lenses. The arithmetic means of the 3 different fields was used for statistical analysis. The mean and standard deviation of the number of labeled cells with BrdU marker in A, B and C groups using previous partial corneal desepithelization technique and SF at temporal, central and nasal area were respectively 15.07 ± 12.18; 5.53 ± 5.11; 1.80 ± 2.37 in group A, 11.17 ± 11.15; 8.50 ± 7.18; 9.67 ± 9.09 in group B e 12.34 ± 9.02; 15.67 ± 5.84; 3.78 ± 2.80 in group C. Using the same technique and MMC the results were: 6.60 ± 5.33; 3.80 ± 6.36 e 1.93 ± 2.14 in group A, 3.09 ± 2.40; 4.34 ± 2.97 e 2.59 ± 0.63 in group B e 5.83 ± 8.55; 6.17 ± 1.55; 4.42 ± 2.63 in group C. For the corneal desepithelization surgical technique after SF application, the results were: 13.80 ± 8.51; 7.47 ± 3.79; 3.07 ± 1.55 in group A, 5.67 ± 4.49; 4.75 ± 2.13; 3.84 ± 3.45 in group B e 6.78 ± 3.35; 11.67 ± 2.97; 3.78 ± 1.39 in group C. When MMC was applied in the same surgical technique, the results were: 8.73 ± 7.35; 7.60 ± 6.88; 3.27 ± 1.85 in group A, 1.75 ± 0.78; 3.92 ± 3.06; 2.67 ± 1.98 in group B e 2.83 ± 3.54; 2.50 ± 2.08; 8.75 ± 9.62 in group C. The difference between MMC and SF were statistically significant at central and temporal areas when previous partial corneal desepithelization was performed in all groups. There was no difference between drugs when corneal desepithelization technique was used after solution application. There was no difference between differentiation cells in the slides labeled with AE1, AE3 and AE5 when drugs or surgical technique were analysed. These results suggests that 0,02% MMC applied for 3 minutes at the episclera, acts only at the exposed cells and have late effect under the cell proliferation, when the previous partial desepithelization surgery was applied
