RESUMO

Sugarcane is one of species more cultivated in whole world, reaching more than 80 countries. With the biotechnological progresses, important tools appear for genetic breeding of cane. The production of transgenic plants is very important for clonal propagation species as sugarcane. But plant transformation is dependent of in vitro cultivation and, among their techniques it is somatic embryogenesis, the object of present study. For embryogenesis induction were evaluated effects of growth regulator 2,4-D in followed concentrations: 0, 1, 3, 6 and 9 mg.L-1. During multiplication was tested 1, 3 and 6 mg.L-1 of 2,4-D. In maturation and germination of callus were evaluated presence and absence of light, and the presence or sucrose absence in the medium of cultivation. It was verified that the conditions of the plant in field affect the surface sterilization process directly. The auxin is very important for embryogenic callus induction, however there wasnt significant difference among different concentrations of growth regulator. During the multiplication it was observed that the interaction among 2,4-D and the genotype was significant. Therefore, it was made the unfolding of analysis. The 3 mg.L-1 of 2,4-D treatment was the most efficient for callus multiplication of clone RB835486. In the clones RB855536 and RB928064 the multiplication of embryogenic callus was inversely proportional to the concentration of 2,4- D in the medium. The presence or sucrose absence in the culture medium didn t influence the germination of the somatic embryos significantly. The same was observed in the callus that were maintained under light (clear) or in the darkness during the 30 days of cultivation. In spite of the limited number of regenerate plantlets, the process of plantlets acclimatization was made with success, because 59% of survivors were obtained.
