Genetic diversity of Phaeoisariopsis griseola isolates revealed by molecular markers / Análise da diversidade genética de isolados de Phaeoisariopsis griseola por meio de marcadores moleculares

RESUMO

The angular leaf spot, caused by Phaeoisariopsis griseola, also known as Pseudocercospora griseola (Sacc.) Crous &U. Braun, is one of the most important diseases in the bean plant, causing serious damages to the production. The most efficient cost strategy for the management of plant diseases is the use of resistant cultivars. However, studies related to the genetic diversity of P. griseola have been demonstrating great genetic variability in this fungus, what has been hardening the achievement of those cultivars. Therefore, the aim of this work was to verify if the techniques of ERIC-PCR, BOX-PCR, ISSR-PCR and PCR- RFLP of the rDNA ITS region are able to detect the genetic diversity of the P. griseola isolates from Goiás, Minas Gerais, Espírito Santo and Paraná states. The total DNA of the 29 P. griseola isolates were extracted and, later, amplified using specific primers for ERIC, BOX, ISRR and ITS sequences. The cleavages of the ITS region with the restriction enzymes AluI, HaeIII, HhaI, HpaII, MboI, MspI e RsaI produced identical restriction profiles for all the isolates, and, therefore, the ITS sequence under used conditions supply markers that could differ the isolates of P. griseola. Through the grouping analysis of the results obtained by the BOX-PCR, ERIC-PCR and ISSR-PCR techniques, 2, 5, and 28 genotypes were revealed, respectively, among the isolates studied. Those results showed that BOX-PCR and ERIC-PCR techniques were less efficient to detect the genetic variability of P. griseola isolates. The best results were obtained using the ISSR- PCR technique, revealing high capacity in the polymorphism detection among the isolates. The genetic diversity observed can help in the development of appropriate strategies to obtain resistant bean plant varieties to this fungus.
