RESUMO

Bean golden mosaic virus in Brazil has not been transmitted mechanically by investigators that have tried it. Other related viruses associated with similar diseases present in Central America, Porto Rico, and Colombia have been transmitted by routine methods of mechanical inoculation without great difficulty. This apparent discrepancy led the writer to start a series of tests aiming at developing a teshnique that wold permit mechanical transmission of the Brazilian virus. Several approaches were tried ? (1) variations in the source of inoculun: (a) simple collected from several regions in the State of São Paulo and from other parts of Brazil; (b) inocula collected from different donor plant species; (c) inocula prepared at different concentrations. (2) Different methods of extraction and use of various extracting solutions: (a) use of virus activators; (b) use of inhibitors of nucleases and proteases or of other substances employed to preserve the integrity of the viral particles; (c) extraction of the inoculum in different solutions and combination at inoculation time or through two successive inoculation; (d) extration after freezing the samples. (3) different methods of inoculations: (a) rubbing on both sides of the leaves; (b) pin pricks on leaves and stems; (c) repeated inoculation of same leaves wit same or different preparations...
