RESUMO

This project analyzed the effect of the opaque ? 2 gene and growth regulators upon the degradation of endosperm reserves during the early seedling growth of maize cv. Maya. Grains of opaque ? 2 maizw were morphologically didtinct, generally lighter with a large scutellum and showed a higher total protein content in both the embryo and endosperm. For both types protein content was positively correlated with grain weight though for heavier grain differences in protein content were greatest in the endosperm, whereas in lighter grain differences were greatest in the germ. Analysis of protein composition revealed that albumin, globulin and glutelin were increased in opaque-2 grain whereas zein was considerably reduced. In spite of initial faster imbibition rates in opaque-2 maizeno differences were discernible in initial plant growth, which was accompanied by a rapid decrease in endosperm dry matter content and protein in both forms, though the latter was utilized relatively faster. Greater quantities of dry matter and protein were translocated to the shoot, particularly the first leaf lamina which expanded rapidly from day 5. In absolute terms degradation was more rapid in the most abundant protein classes, however zein and salt soluble fractions were utilized relatively faster in normal maize when compared with opaque-2 maize where glutelin degradation was relatively greater ...Note: The complete abstract is available with the full electronic digital thesis or dissertations
