Estudo quimico e avaliação antiproliferativa de diferentes especies do genero Mikania

RESUMO

The genera Mikania possess over 430 species and is distributed throughout the tropical regions of Africa, Asia and South America. Previous chemical and pharmacological results with two Mikania species prompted us to evaluate Mikania argyreiae, M. glomerata (Iguape), M. glomerata (Picinguaba), M. laevigata (Iguape) and M. hirsutissima hydroalcoholic, dichloromethane and dichloromethane fractions activity by in vitro antiproliferative assay with neoplasic human cells, in order to isolate and identify the active compounds responsible for the pharmacological activity under study. The extracts were prepared from dry grinded plants. The evaluation of the chemical composition was carried out by chromatographic methods. The antiproliferative assay were carried out in 96 well plates employind four extract concentration (0,25; 2,5; 25 and 250 µg/mL). The dosage of proteins was carried through by coloration with sulforrodamine B and espectrofotometric reading in 560 nm. The phytochemistry studies had disclosed small variations in the chemical composition between the species, with prominence for the M. hirsutissima of which was isolated the pimaridienic acid, not described previously for this species and absente in the others. This acid was isolated by dry column and classic chromatography and identified by spectroscopic methods using 13CNMR and 1HNMR techniques, mass and infrared spectroscopy. The antiproliferative activity evaluation demonstrated that the M. glomerata (Iguape and Picinguaba) extracts presented different profiles between the same species from different origins. M. hirsutissima crude extract and polar fractions presented inhibited growth at the highest concentration and pimaridienic acid presented a different antiproliferativa activity compared to caurenoic and cupressenic diterpenic acids, previously tested, demonstrating an intrinsic relationship with the chemical structure.
