RESUMO

The fruticultura of tempering climate is an activity of great partner-economic importance in Brazil, being that the biggest area of production is located in the South Region of the country, and the State of Santa Catarina is responsible for about 50% of offers national. The consumption of the apple comes increasing and with the fast increase of the area of culture of the apple tree, it had an addition of resistant to biotic and abiotic stresses. To keep the culture of the apple tree in level of desired productivity, the use of biotechnological techniques for attainment of dumbs of proven genetics-sanitary quality, of the new varieties, the production of basic first plants of better quality and productivity, they are some of the important conditions for the apple tree. The objective of this work was to develop regeneration protocols in vitro for apple tree (Malus domestica Borkh) cv. Galaxy, of organogenesis direct, for propagation studies in vitro and genetic improvement. For used as internodal explants and leaves where they had been introduced in cultured MS medium, with BAP, TDZ and ANA. In the establishment in vitro the applied treatments of asepsis had been little efficient, presented contamination indices that had varied between 5-40%. In the multiplication process the concentration of 4,44 M of BAP was most effective for multiplication and production of shoots. Regarding organogenesis from leave explant, it could be observed that the type of explant, that is, the cut of the limb was favorable to the process of regeneration of shoots. However, the explant entire leaf presented the best ones resulted, where a bigger number of regenerated explants was gotten. The concentration of 9,1 M of TDZ added to the medium of culture, was the concentration that presented the favorable results the regeneration of shoots. However, for folial disks, the concentration that presented resulted superior was 13,62 M of TDZ. The results of the histological analyses allow to infer that organogenesis of the shoots, had direct origin of cells of mesophyll, or either, cells of parenchyma, however we cannot affirm that all the adventitious buds had originated, therefore observed it presence of callus in the explants in elapsing of the experiments.
